11. will not be applied to seal the plate Multi well plates not sealed properly Ensure that the sealing film or tape used is rated to withstand up to 120 C Problems during Run Symptom Possible Cause If changes are made to a program and the program is re saved with the same name the changes will not be sent to the main computer unless the program is re loaded To check the Running program does not seem to match parameters actually being read by the cycler saved program during the run open the Run Options Menu and select View Current Program Settings If the settings show the original program stop the running program and Load Another Program from the Run Options Menu Ensure that there is some kind of tube or plate in the cycler If no reaction vessel is present there is no buffer between the lid while it is heating and the block while it is chilling and the lid will not be able to reach the correct temperature The threshold for the heated lid is 25 1 C If a step is programmed to 25 0 C or lower the heated lid turns off After the machine finishes the 25 0 C cycle it will go into lid heating mode again which means the block will go Lid heating appears in the middle of a cycle down to 4 C to hold the tubes while the lid turns back on and heats up Once the lid is fully heated the block temperature will rise from 4 C to the next programmed temperature To avoid this delay ensure that all
