11. 2479 Strassberg i Germany Tel 49 743493640 Fax 49 7434936440 ij www aid diagnostika com SECTION 3 The AID EliSpot Reader in the Lab 3 1 Set up the system for routine use This section explains how to set up and calibrate the AID EliSpot Reader System for use with one or more types of plates create and manage count settings set the camera For this you need to be able to do all that was explained in the previous section You should also be reasonably acquainted with the scientific background of the experiments You need to e recognize common artifacts in EliSpot e decide which spots you want to include or exclude in the analysis e decide on issues of quality management Getting good pictures of each well of an EliSpot plate is essential since the actual plate will change over time and may fade considerably Once a plate document has been stored on the hard disk or other media it can be reanalyzed again and again without loss of information First of all the plate needs to be in focus Focus is adjusted in the calibration dialog box with the buttons on the very right of the box Note that in older systems focusing is done with the up and down switch on the Reader Unit The optical set up of the system has a very good depth of focus so that minor variations are not an issue However if a particularly bent plate is to be read special precautions must be taken To focus the Reader insert plate read plate then choose a well ne
